Biotinylation of (a) sulfhydryl containing peptides and proteins; (b) terminal sulfhydryl modified DNA at acidic to neutral pH. This is the only comercially available sulfhydryl reactive biotinylation reagent with hydrophilic spacer arm and maleimide functionality. Compounds like biotin-HPDP and biotin-BMCC impart detrimental properties when labeled due to the presence of both a very highly hydrophobic spacer and reactive functionality. Spacer length is ideal for rapid and tight avidin/streptavidin binding. Biotin-amide carbonyl to terminal maleimide is 24.9A (21 atoms). Hydrophilic spacer arm eliminates or minimizes non-specific binding that causes aggregation and precipitation problems, which commonly occur when labeling antibodies and other biological materials. Hydrophilic dPEGTM spacer is non-immunogenic. Ultra high purity. |

Note: Greg has protocols using biotin-BMCC for the biotinylation of (a) sulfhydryl containing peptides and proteins on page 360; and (b) terminal sulfhydryl modified DNA on page 660. They are equally applicable to this reagent, however, with the major additional advantage of our reagent's enhanced water and organic solubility. It is readily soluble in DMF, DMAC (N, N-dimethylacetamide), methylene chloride, and in water. |
